A genome-wide ordered-subset linkage analysis for rheumatoid arthritis
Rheumatoid arthritis (RA) is a chronic, complex autoimmune inflammatory disorder with poorly known etiology. Approximately 1% of the adult population is afflicted with RA. Linkage analysis of RA can be complicated by the presence of phenotypic and genetic heterogeneity. It is shown that the ordered-subset analysis (OSA) technique reduces heterogeneity, increases statistical power for detecting ...
متن کاملOrdered subset analysis in genetic linkage mapping of complex traits.
Etiologic heterogeneity is a fundamental feature of complex disease etiology; genetic linkage analysis methods to map genes for complex traits that acknowledge the presence of genetic heterogeneity are likely to have greater power to identify subtle changes in complex biologic systems. We investigate the use of trait-related covariates to examine evidence for linkage in the presence of heteroge...
متن کاملAnalysis of matched case-control studies.
There are two common misconceptions about case-control studies: that matching in itself eliminates (controls) confounding by the matching factors, and that if matching has been performed, then a “matched analysis” is required. However, matching in a case-control study does not control for confounding by the matching factors; in fact it can introduce confounding by the matching factors even when...
متن کاملBayesian analysis of case-control studies.
A Bayesian approach to the estimation of an odds ratio from case-control data is considered. The exact posterior density of the odds ratio and its moments are derived. A log-normal approximation to the density is shown to be adequate for practical purposes. Mechanisms for setting prior parameters are discussed and some examples are presented.
